KGX101 is an engineered prodrug of the cytokine interleukin-12 (IL-12), developed as a recombinant fusion protein. It consists of human IL-12 fused to an antibody Fc region and masked with tumor-specific protease-cleavable linkers. This design prolongs serum half-life and restricts activation to the tumor microenvironment, where local proteases cleave the linker to release active IL-12. The released cytokine stimulates immune cells such as T lymphocytes and NK cells within tumors, promoting anti-tumor immunity while minimizing systemic toxicity. Preclinical studies have demonstrated potent anti-tumor activity and favorable pharmacokinetics. Clinical trials are ongoing for advanced or metastatic solid tumors as monotherapy or in combination with PD-L1 inhibitors[1][2][3][4][5].
