KGX101 + envafolimab is an investigational combination therapy comprising two biologic agents for cancer treatment: - **KGX101** is a recombinant interleukin-12 (IL-12) Fc fusion protein prodrug designed for intravenous injection. It aims to enhance anti-tumor immune responses by delivering IL-12, a cytokine that stimulates immune cell activity, particularly T cells and natural killer (NK) cells. - **Envafolimab** is a subcutaneously administered, humanized single-domain anti-PD-L1 antibody fused to an IgG1 Fc fragment (also known as KN035). It functions as a checkpoint inhibitor targeting the PD-L1 pathway, thereby enhancing T cell-mediated immune responses against tumors. The combination is under Phase 1 clinical investigation for use in advanced or metastatic solid tumors, aiming to synergize immune activation from both IL-12 (KGX101) and PD-L1 inhibition (envafolimab). KGX101 is developed by KangaBio/Shanghai Kangkang Biopharmaceutical, while envafolimab was originally developed collaboratively by Alphamab and 3D Medicines.
