Overview

KGX105 is a tumor-specific protease-sensitive bispecific antibody (BsAb) designed as a conditionally active prodrug T-cell engager targeting both CD3 on T cells and EGFR (epidermal growth factor receptor) on tumor cells. Developed by KangaBio (Shanghai Kangkang Biopharmaceutical Co., Ltd.), this agent aims to induce potent anti-tumor cytotoxicity by redirecting T lymphocytes to kill EGFR-expressing cancer cells while minimizing off-tumor toxicity and systemic cytokine release. The molecule is engineered to be activated specifically in the tumor microenvironment via protease cleavage, thereby improving safety and specificity compared to conventional CD3-bispecific antibodies. Preclinical studies have demonstrated significant anti-tumor efficacy in models of colorectal cancer and other solid tumors[1][2][3][4][8].
