KH-3 is a small-molecule inhibitor of the RNA-binding protein **HuR** (ELAV-like protein 1; ELAVL1). HuR is a post-transcriptional regulator that binds to AU-rich elements (AREs) within the 3' untranslated regions of target mRNAs, enhancing their stability and translation. By disrupting HuR's ability to bind its targets, KH-3 promotes the decay of specific transcripts involved in inflammation and metabolic wasting. In preclinical models of **allergic asthma**, KH-3 has been shown to suppress the expression of the master transcription factor **GATA3** and downstream Th2 cytokines (IL-4, IL-5, IL-13), thereby reducing airway inflammation and hyperresponsiveness. Additionally, KH-3 is being investigated for its potential to reverse adipose tissue wasting in **pancreatic cancer cachexia** by restoring the expression of adipogenesis regulators such as **Pparg** and **Cebpa**.
