KH-801 is a differentiated humanized IgG1 monoclonal antibody targeting CD24, developed for the treatment of advanced solid tumors. It functions as a CD24 antagonist and exerts anti-tumor effects by promoting phagocytosis of tumor cells through both Fc-mediated immune effector mechanisms (including ADCC and ADCP) and blockade of the CD24/Siglec10 axis. This dual mechanism enhances macrophage-mediated tumor cell clearance, increases T cell infiltration into tumor tissues, and promotes elimination of M2 tumor-associated macrophages (TAMs), potentially transforming immunologically "cold" tumors into "hot" ones. Preclinical studies have demonstrated dose-dependent anti-tumor efficacy in multiple mouse xenograft models with good tolerance observed in non-human primate toxicity studies. As of 2024, KH-801 is being evaluated in Phase 1 clinical trials for safety, tolerability, pharmacokinetics, immunogenicity, and preliminary efficacy in patients with advanced solid tumors[1][2][4].
