KH813 is a recombinant humanized bifunctional fusion protein developed by Chengdu Kanghong Biotech for the treatment of advanced solid tumors. It is designed to simultaneously target and inhibit the Programmed cell death 1 ligand 1 (PD-L1) and Transforming growth factor beta (TGF-β) signaling pathways. The molecule typically consists of an anti-PD-L1 monoclonal antibody fused to the extracellular domain of the TGF-β receptor II (TGF-βRII), which acts as a trap to neutralize TGF-β. By blocking PD-L1, KH813 prevents the suppression of T-cell activity, while the sequestration of TGF-β helps to reverse the immunosuppressive and pro-fibrotic effects within the tumor microenvironment. This dual-targeting approach is intended to overcome resistance to standard immune checkpoint inhibitors and enhance anti-tumor immune responses.
