KH815 is a novel dual-payload antibody-drug conjugate (ADC) developed by Chengdu Kanghong Biotech for the treatment of advanced malignant solid neoplasms. It targets the trophoblast cell surface antigen 2 (TROP2) using a humanized IgG1 antibody and delivers two cytotoxic payloads—a topoisomerase I inhibitor and an RNA polymerase II inhibitor—enabling simultaneous inhibition of tumor cells at both the DNA and RNA levels. This dual mechanism aims to overcome drug resistance, reduce P-glycoprotein (P-gp) and HSP70 protein expression, and improve chemotherapy sensitivity. Preclinical studies have shown that KH815 exhibits dose-dependent tumor growth inhibition in various xenograft models, including those resistant to other ADCs such as Trodelvy. The drug has demonstrated superior antitumor activity compared to single-payload ADCs with an acceptable safety profile in non-human primates. As of April 2025, KH815 is entering phase 1 clinical trials for advanced solid tumors[1][4][5][7].
