KHK2866 is a recombinant, humanized, non-fucosylated monoclonal antibody that targets heparin-binding epidermal growth factor-like growth factor (HB-EGF). By binding to HB-EGF, KHK2866 blocks its interaction with EGF receptors and neutralizes its activity. This mechanism is intended to inhibit tumor cell proliferation and survival in cancers where HB-EGF plays a role. The drug was developed as an antineoplastic agent for the treatment of advanced solid tumors, including ovarian cancer. In clinical studies, it demonstrated dose-dependent pharmacokinetics and reduced serum free HB-EGF levels but was associated with hypersensitivity reactions and neurotoxicity at higher doses. Development has been discontinued following phase 1 trials due to safety concerns and lack of further efficacy data[1][2][3][5][7].
