KHK4577 is an orally active small molecule antagonist of the histamine H4 receptor (H4R) developed by Kyowa Kirin for the treatment of atopic dermatitis. The histamine H4 receptor is a G protein-coupled receptor primarily expressed on hematopoietic cells, including mast cells, eosinophils, dendritic cells, and T cells, where it mediates chemotaxis and the production of pro-inflammatory cytokines. By selectively antagonizing H4R, KHK4577 aims to reduce the pruritus (itching) and underlying skin inflammation associated with atopic dermatitis. The compound has been evaluated in Phase 2 clinical trials in Japan to assess its efficacy and safety in patients with moderate-to-severe disease.
