KHN707 is an orally bioavailable, highly selective thyroid hormone receptor beta (THR-β) agonist developed by Chengdu Kanghong Pharmaceutical Group. THR-β is the primary isoform of the thyroid hormone receptor expressed in the liver, where it regulates key metabolic pathways including lipid metabolism and bile acid synthesis. By selectively targeting the beta isoform, KHN707 aims to reduce hepatic fat accumulation and inflammation associated with metabolic dysfunction-associated steatohepatitis (MASH), formerly known as non-alcoholic steatohepatitis (NASH), while minimizing systemic side effects mediated by the THR-alpha isoform, such as cardiotoxicity and bone loss. It is currently in early-phase clinical development, with studies evaluating its safety, tolerability, and pharmacokinetic profile in healthy volunteers.
