KI16425 is a small molecule lysophosphatidic acid (LPA) receptor antagonist with subtype selectivity for LPA1 and LPA3, demonstrating moderate activity against LPA2. It inhibits LPA-induced cellular activities such as calcium signaling, DNA synthesis, cell migration, and survival in various cancer and inflammatory models. Mechanistically, KI16425 blocks LPA-mediated interactions and signaling that drive inflammation, tumor progression, and metastasis, and it can suppress inflammatory cytokine production and tumor cell proliferation.
