Overview

KI16425 is a small molecule lysophosphatidic acid (LPA) receptor antagonist with subtype selectivity for LPA1 and LPA3, demonstrating moderate activity against LPA2. It inhibits LPA-induced cellular activities such as calcium signaling, DNA synthesis, cell migration, and survival in various cancer and inflammatory models. Mechanistically, KI16425 blocks LPA-mediated interactions and signaling that drive inflammation, tumor progression, and metastasis, and it can suppress inflammatory cytokine production and tumor cell proliferation. Preclinical studies suggest potential utility in modulating immune response, reducing inflammation, and inhibiting cancer metastasis[1][2][3][6][7].
