The killed Mia PaCa-2 cells vaccine is an allogeneic whole-cell cancer vaccine derived from the Mia PaCa-2 human pancreatic carcinoma cell line. The vaccine consists of Mia PaCa-2 cells that have been inactivated, typically through lethal irradiation, to prevent their proliferation while preserving the structural integrity of their tumor-associated antigens (TAAs). The mechanism of action involves the presentation of a broad spectrum of pancreatic cancer-specific antigens to the host's immune system, thereby stimulating a polyclonal T-cell and B-cell response against the patient's own tumor cells. This approach is often enhanced by genetic modification of the cell line to express immunostimulatory molecules such as GM-CSF, IL-2, or alpha-1,3-galactosyl epitopes (as seen in algenpantucel-L), which facilitate the recruitment and activation of dendritic cells and other antigen-presenting cells. While primarily investigated for the treatment of pancreatic adenocarcinoma, clinical development has faced challenges in achieving significant overall survival benefits in large-scale trials.
