**KIN1148** is a small-molecule vaccine adjuvant discovered by Kineta using their proprietary AViid™ platform. It directly binds and activates retinoic acid-inducible gene I (RIG-I), inducing RIG-I self-oligomerization and downstream signaling through IRF3 and NFκB pathways in an RNA- and ATP-independent manner, leading to expression of immunomodulatory cytokines and chemokines without strong type I interferon induction. This enhances humoral and cellular immune responses to influenza vaccines, providing dose-sparing effects (>15-fold), neutralizing antibodies, T cell activation, and protection against lethal H1N1 and H5N1 challenges in preclinical models while promoting balanced Th2 and immunoregulatory responses to mitigate immunopathology. Developed in collaboration with NIH and University of Washington, it is formulated in liposomes for intramuscular delivery.[1][2][3][4]
