KIO-101

KIO-101 is a small molecule inhibitor of dihydroorotate dehydrogenase (DHODH), developed as a topical eye drop for the treatment of ocular inflammation associated with autoimmune diseases, including the ocular presentation of rheumatoid arthritis and dry eye disease. As a DHODH inhibitor, KIO-101 reduces T-cell proliferation and proinflammatory cytokine release, targeting local immune responses in the eye without systemic side effects typical of other anti-inflammatory drugs. The drug is being developed by Kiora Pharmaceuticals and has demonstrated safety, tolerability, and anti-inflammatory activity in Phase 1 clinical studies. It is currently in Phase 2 trials for patients with ocular manifestations of autoimmune diseases such as rheumatoid arthritis, psoriatic disease, systemic lupus erythematosus, and fibromyalgia[1][3][4][6][7][8].
