Overview

KIR-CAR T-cells are a novel chimeric antigen receptor (CAR) T-cell therapy platform developed by Verismo Therapeutics, utilizing a multichain design inspired by the natural biology of natural killer (NK) cells. Unlike conventional CAR T-cells that typically use a single-chain CD3ζ signaling domain, KIR-CAR T-cells incorporate killer immunoglobulin-like receptors (KIR) and the DAP12 signaling adapter protein. This dual-chain architecture creates a regulated coupling/decoupling mechanism that acts as a natural on/off switch, allowing the T-cells to enter a resting state when not engaged with tumor antigens. This design is intended to reduce T-cell exhaustion and enhance persistence within the challenging microenvironment of solid tumors. The lead candidate, SynKIR-110, is an autologous cell therapy targeting mesothelin-expressing cancers, including mesothelioma, ovarian cancer, and cholangiocarcinoma.
