The Kir4.1 inhibitor program, developed by the Warren Center for Neuroscience Drug Discovery (WCNDD), is a preclinical-stage drug discovery effort focused on identifying small-molecule inhibitors of the Kir4.1 (KCNJ10) inward rectifier potassium channel. Kir4.1 is an astrocyte-specific potassium channel that plays a critical role in maintaining potassium homeostasis in the central nervous system. Research suggests that increased expression or activity of Kir4.1 in the lateral habenula is associated with the pathophysiology of Major Depressive Disorder (MDD). By inhibiting Kir4.1, these small molecules aim to normalize neuronal firing patterns and astrocyte-neuron interactions, potentially offering a novel therapeutic approach for patients with MDD.
