Overview

KISIMA-OVA is a therapeutic cancer vaccine candidate developed using the KISIMA platform technology, originally by AMAL Therapeutics (now part of Boehringer Ingelheim). It is a recombinant fusion protein designed to elicit a robust and targeted cytotoxic T-cell response. The construct comprises three functional components: a cell-penetrating peptide (CPP) derived from the ZEBRA protein for efficient intracellular delivery, a multi-antigenic cargo containing the model antigen ovalbumin (OVA), and a Toll-like receptor (TLR) peptide agonist that serves as a built-in adjuvant. By delivering antigens directly to dendritic cells and simultaneously providing maturation signals, KISIMA-OVA promotes the expansion of antigen-specific CD8+ T cells. While primarily used as a proof-of-concept tool in preclinical oncology models, it demonstrates the modularity and immunogenicity of the KISIMA platform, which is being applied to clinical candidates such as ATP128 for colorectal cancer.
