KP54 (kisspeptin-54), also known as metastin, is a 54-amino acid peptide derived from the proteolytic cleavage of the KISS1 metastasis suppressor protein. It is the primary endogenous ligand for the G protein-coupled receptor KISS1R (also known as GPR54). KP54 is primarily recognized for its potent ability to suppress the metastatic colonization of cancer cells, particularly in melanoma and breast cancer, without affecting the growth of the primary tumor. While its canonical mechanism involves the activation of KISS1R, research from the University of Kansas Medical Center suggests that KP54 and other KISS1-derived peptides may also suppress metastasis through KISS1R-independent pathways, involving the inhibition of cell motility and the upregulation of mitochondrial oxidative phosphorylation. Additionally, KP54 plays a critical role in the hypothalamic-pituitary-gonadal axis by stimulating the secretion of gonadotropin-releasing hormone (GnRH).
