A kisspeptin-9 analogue is a synthetic peptide agonist of the kisspeptin receptor (KISS1R, also known as GPR54). It is a shortened, modified version of the endogenous kisspeptin peptide (typically a nonapeptide) designed to possess enhanced stability, potency, and water solubility compared to native kisspeptin-10 or kisspeptin-54. Developed and evaluated in clinical physiology studies by Imperial College London, kisspeptin-9 analogues are being investigated for the treatment of fertility disorders. By binding to KISS1R on gonadotropin-releasing hormone (GnRH) neurons in the hypothalamus, these analogues stimulate the pulsatile release of GnRH, which subsequently triggers the secretion of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) from the anterior pituitary, thereby regulating the hypothalamic-pituitary-gonadal axis.
