Overview

KIT2014 is an inhaled, cell-permeable cAMP-modulating peptide therapy being developed by Kither Biotech for cystic fibrosis and other obstructive airway diseases. It acts as a PI3K gamma mimetic peptide that disrupts the interaction between phosphoinositide 3-kinase gamma (PI3Kγ) and protein kinase A (PKA), leading to functional inhibition of type 3 and 4 phosphodiesterases (PDE3/4), increased intracellular cAMP, and downstream enhancement of cystic fibrosis transmembrane conductance regulator (CFTR) channel gating and stability at the plasma membrane.[1][7] Local cAMP elevation in bronchial epithelial cells improves CFTR-mediated chloride transport, while in airway smooth muscle and immune cells it promotes bronchodilation and reduces neutrophil-driven inflammation, thereby addressing mucus obstruction, airway inflammation, and bronchoconstriction when used as an add-on to existing CFTR modulators.[1][2][4][8] KIT2014 is delivered by inhalation (via nebulizer) and has received EMA orphan drug designation for cystic fibrosis; it is in early clinical development with an initial Phase 1 trial in healthy volunteers and is also being explored preclinically for COPD and asthma.[2][3][8][10]
