KITE-585 is an investigational autologous anti-BCMA chimeric antigen receptor (CAR) T-cell therapy developed for the treatment of relapsed/refractory multiple myeloma. It is engineered from a patient’s own T cells to express a chimeric antigen receptor containing a fully human monoclonal antibody targeting B-cell maturation antigen (BCMA), a protein expressed on malignant plasma cells and some B-cell lineages, but not on other tissues. KITE-585 incorporates a CD28 costimulatory domain and a proprietary linker designed to optimize T-cell activation, expansion, and function without tonic signaling. It has demonstrated specificity and selectivity to BCMA-expressing targets and polyfunctional activation in preclinical studies. In a Phase 1 clinical context, KITE-585 showed a manageable safety profile but minimal clinical efficacy due to limited CAR T-cell expansion in vivo. The therapy was developed by Kite Pharma, a Gilead company, and has been evaluated primarily for patients with multiple myeloma refractory to multiple prior lines of therapy[1][2][3][5][7][8][9].
