Overview

KJ-C2320 is an allogeneic chimeric antigen receptor (CAR) T-cell therapy developed by CARsgen Therapeutics. It targets CD38, a cell surface protein highly expressed in certain hematologic malignancies. The therapy is based on CARsgen’s THANK-uCAR platform, which modifies donor-derived T cells to improve their expansion and persistence while minimizing graft-versus-host disease (GvHD) and host-versus-graft response (HvGR). This is achieved by disrupting the genomic loci encoding the T-cell receptor (TCR) and beta-2 microglobulin (B2M), eliminating surface expression of both TCR and human leukocyte antigen class I (HLA-I). To further protect these engineered cells from natural killer (NK) cell-mediated rejection, the THANK-uCAR platform arms them with a chimeric antigen receptor that recognizes NKG2A, thereby hindering NK cell attacks. KJ-C2320 is currently being evaluated in an investigator-initiated Phase 1 trial for relapsed/refractory acute myeloid leukemia in China[1][4][6].
