Overview

KK2269 is a novel bispecific antibody developed using Kyowa Kirin's REGULGENT technology. It targets both CD40, an immune co-stimulatory receptor, and epithelial cell adhesion molecule (EpCAM), a tumor-associated antigen. By engaging CD40 and EpCAM simultaneously, KK2269 is designed to activate dendritic cells within the tumor microenvironment while directly targeting tumor cells expressing EpCAM. This dual mechanism aims to enhance T-cell priming and activation against tumors, potentially increasing antitumor immune responses. Preclinical studies have shown that combining KK2269 with chemotherapy (such as docetaxel) results in synergistic effects on tumor reduction and increased activation of dendritic cells. The drug is currently being evaluated in Phase I clinical trials for advanced or metastatic solid tumors—including non-small cell lung cancer, gastric adenocarcinoma, gastroesophageal junction adenocarcinoma, and esophageal adenocarcinoma—administered intravenously as monotherapy or in combination with docetaxel[1][4][5][6][7][8][9].
