KK2845 is an antibody-drug conjugate (ADC) developed by Kyowa Kirin targeting T-cell immunoglobulin mucin-3 (TIM-3). It is designed to treat hematologic malignancies, specifically acute myeloid leukemia (AML), by targeting TIM-3 expressed on leukemic stem cells and blasts. The molecule consists of a non-fucosylated fully human anti-TIM-3 IgG1 monoclonal antibody, which is engineered for enhanced antibody-dependent cellular cytotoxicity (ADCC), conjugated to a pyrrolobenzodiazepine (PBD) dimer payload (SG3249, also known as tesirine) via a valine-alanine linker. Upon binding to TIM-3 and internalization, the PBD payload is released, leading to DNA cross-linking and cell death. KK2845 is currently being evaluated in Phase I clinical trials for relapsed or refractory AML.
