KK2845_3 is an investigational antibody-drug conjugate (ADC) developed by Kyowa Kirin. It targets T-cell immunoglobulin and mucin-domain containing-3 (TIM-3), a receptor expressed on leukemia stem cells in acute myeloid leukemia (AML). The ADC consists of a fully human IgG1 anti-TIM-3 antibody conjugated to the highly potent DNA cross-linking pyrrolobenzodiazepine (PBD) dimer payload SG3199 via a valine-alanine linker. By binding to TIM-3-expressing cells, the drug delivers the cytotoxic PBD dimer directly to malignant cells, inducing cell death through DNA cross-linking. Its primary indication under investigation is relapsed or refractory acute myeloid leukemia[4][5].
