Overview

KK6483C is an investigational small molecule catechol-O-methyltransferase (COMT) inhibitor being developed by Kyowa Kirin for the treatment of Parkinson's disease. COMT inhibitors are used as adjuncts to levodopa therapy to prevent the peripheral degradation of levodopa into 3-O-methyldopa. By inhibiting this metabolic pathway, KK6483C increases the bioavailability and extends the plasma half-life of levodopa, allowing more of the drug to cross the blood-brain barrier and be converted into dopamine in the central nervous system. This mechanism is intended to reduce "off" periods and improve motor control in patients experiencing motor fluctuations. KK6483C has been evaluated in Phase 1 clinical trials in Japan, including pharmacokinetic studies assessing its safety and interaction when administered alongside standard triple therapy (levodopa/carbidopa/entacapone).
