Overview
KL-140 (also known as A140 and branded as Datalai) is a recombinant anti-epidermal growth factor receptor (EGFR) monoclonal antibody that serves as a biosimilar to cetuximab. Developed by Sichuan Kelun Pharmaceutical Research Institute and Sichuan Kelun Botai Biomedicine, the drug acts as an EGFR antagonist, binding specifically to the extracellular domain of the EGFR (ErbB1). This binding prevents the interaction between the receptor and its endogenous ligands, such as epidermal growth factor (EGF) and transforming growth factor-alpha (TGF-alpha), thereby inhibiting downstream signaling pathways involved in cell proliferation, survival, and angiogenesis. KL-140 is primarily being developed for the first-line treatment of RAS wild-type metastatic colorectal cancer (mCRC) and is also being investigated for use in head and neck squamous cell carcinoma (HNSCC).
