KL-A289 is an orally bioavailable small molecule antagonist of the adenosine A2A receptor (A2AR), developed by Anyuan Pharmaceutical Technology (Shanghai) Co., Ltd. for the treatment of advanced solid tumors. In the hypoxic tumor microenvironment, adenosine levels are significantly elevated, leading to the activation of A2AR on immune cells such as T lymphocytes and natural killer (NK) cells. This activation triggers an immunosuppressive signaling cascade that inhibits immune cell proliferation and cytokine production, allowing the tumor to evade immune detection. By selectively antagonizing A2AR, KL-A289 prevents adenosine binding and restores the anti-tumor activity of the immune system. It is currently undergoing Phase 1 clinical evaluation as a monotherapy and potentially in combination with checkpoint inhibitors.
