KL060332 is an investigational small molecule hepatitis B virus (HBV) capsid assembly modulator (CAM) developed by Sichuan Kelun Botai Biopharmaceutical. It specifically targets the HBV core protein (HBcAg), a multifunctional protein critical for viral capsid assembly, pregenomic RNA (pgRNA) encapsidation, and viral DNA synthesis. By binding to the core protein, KL060332 induces the formation of aberrant, non-functional capsids or prevents the assembly of capsids altogether, thereby halting the production of new infectious virions. This mechanism is intended to provide a potent antiviral effect in patients with chronic hepatitis B, potentially in combination with nucleos(t)ide analogues. It is currently being evaluated in Phase 1/2 clinical trials in China.
