Drug intelligence / Profile preview
Klebsiella pneumoniae phage P1524 is a novel lytic bacteriophage belonging to the *Przondovirus* genus, isolated from hospital sewage. It exhibits broad-spectrum lytic activity against high-risk, multidrug-resistant, and hypervirulent clones of *Klebsiella pneumoniae* (including ST11, ST14, and ST65 sequence types). The phage combats *K. pneumoniae* infections through a receptor-mediated evolutionary trade-off, where resistance to the phage is driven by a mutation in the host's *wcaJ* gene, leading to capsule loss, impaired biofilm formation, and reduced in vivo virulence. Additionally, phage P1524 encodes several antibacterial effectors, including Orf4, Orf15, and Orf23. Orf4 and Orf23 downregulate key enzymes of the host's 2,3-butanediol fermentation pathway (such as AldB_1, AlsS, and BudC), causing metabolic reprogramming and inhibiting bacterial growth, while Orf23 suppresses biofilm formation. The phage's HHI protein also directly binds to the host's H-NS protein to inhibit bacterial growth [1]. It is being investigated for the treatment of *Klebsiella pneumoniae* infections.
