Overview

**KLF2-overexpressing neoantigen-reactive tumor-infiltrating lymphocytes (TILs)** are a novel engineered cell therapy modality designed to enhance anti-tumor efficacy. Tumor-infiltrating lymphocytes are extracted from a patient's tumor, selectively enriched for those reactive to neoantigens (tumor-specific mutation-derived antigens), and genetically modified to overexpress **KLF2** (Krüppel-like factor 2), a transcription factor that promotes T cell persistence, residency, and stemness while reducing exhaustion. This modification leverages KLF2's role in positively correlating with CD8+ and CD4+ T cell infiltration, dendritic cell activation, and favorable tumor immune microenvironments, as observed in breast cancer and other solid tumors. The therapy aims to improve durable responses in advanced cancers by combining neoantigen specificity with KLF2-mediated T cell fitness enhancement.
