Drug intelligence / Profile preview
KLG-3 is a human IgG4 monoclonal antibody that specifically targets interleukin-13 receptor subunit alpha-2 (IL-13Rα2), a cell-surface receptor overexpressed in various solid tumors, including glioblastoma, melanoma, leiomyosarcoma, and breast cancer, while showing minimal expression in normal tissues. Developed through a collaboration involving Memorial Sloan Kettering Cancer Center, the Tri-Institutional Therapeutics Discovery Institute, and the University of Texas MD Anderson Cancer Center, KLG-3 is designed for theranostic applications. It can be radiolabeled with zirconium-89 (89Zr) for immuno-PET imaging to noninvasively visualize IL-13Rα2-positive tumors, or with therapeutic radionuclides such as lutetium-177 (177Lu) and actinium-225 (225Ac) for targeted radioimmunotherapy. Preclinical studies have demonstrated that KLG-3 exhibits high tumor uptake, favorable pharmacokinetics, and significant antitumor efficacy with minimal off-target toxicity.
