KLG3BBz-huC825 represents a novel theranostic platform utilizing engineered second-generation anti-IL13Rα2-scFv-derived CAR-T cells. These CAR-T cells are designed to target and exert antigen-specific cytotoxicity and cytokine production against Interleukin-13 receptor subunit alpha-2 (IL13Rα2)-expressing cancer cells, such as those found in melanoma. A key feature of this platform is the co-expression of a membrane-bound scFv, huC825, which binds DOTA-haptens with picomolar affinity. This allows for non-invasive, longitudinal tracking of the engineered T cells in vivo using immunoPET with diagnostic radionuclides like [86Y]Y-ABD, providing insights into biodistribution, persistence, and tumor engagement. The platform aims to improve understanding and guidance for next-generation CAR-T cell therapy.
