KLH + Tumor Lysate Pulsed DC Vaccine is an investigational, autologous dendritic cell (DC)-based cancer immunotherapy developed by the H. Lee Moffitt Cancer Center and Research Institute. The vaccine is produced by harvesting a patient's peripheral blood monocytes, differentiating them into immature dendritic cells using GM-CSF and IL-4, and then pulsing them with autologous tumor cell lysate and keyhole limpet hemocyanin (KLH). The tumor lysate provides a broad spectrum of patient-specific tumor-associated antigens, while KLH acts as a highly immunogenic carrier protein and helper antigen to enhance T-cell recruitment and activation. This personalized vaccine is designed to stimulate a robust, tumor-specific T-cell immune response. It is currently being evaluated in Phase 1 clinical trials (e.g., NCT02745756) for the treatment of high-risk neuroblastoma in pediatric patients, often administered following autologous hematopoietic progenitor cell transplant (autoHPCT) to target residual disease.
