Overview

KLH-6-SM is an **experimental vaccine for opioid dependence** composed of a conjugate of keyhole limpet hemocyanin (KLH) and 6-succinylmorphine (6-SM). The vaccine is designed to generate antibodies against morphine and heroin metabolites, thereby sequestering these opioids in the periphery and reducing their entry into the brain. Its mechanism of action is pharmacokinetic antagonism—rather than directly interacting with opioid receptors, it promotes antibody binding to morphine and related compounds, which attenuates both their rewarding (as measured by conditioned place preference) and analgesic effects in animal models. KLH-6-SM has demonstrated high and sustained anti-morphine antibody levels, reduced brain morphine concentrations, and significant attenuation of morphine-induced behavioral effects in rats. Its potential indication is the treatment of **opioid dependence** as an immunotherapy[1].
