Overview

The KLH-adrenomedullin mRNA vaccine is an experimental cancer immunotherapy designed to target adrenomedullin (AM), a regulatory peptide overexpressed in many solid tumors that promotes angiogenesis and tumor growth. The vaccine utilizes in vitro-synthesized mRNA encoding a fusion protein of keyhole limpet hemocyanin (KLH) and adrenomedullin, encapsulated in lipid nanoparticles (LNPs). By acting as a hapten, the KLH component helps break peripheral immunotolerance to the self-antigen adrenomedullin. Administered via intramuscular injection, the vaccine elicits both humoral (anti-AM IgG antibodies) and cellular (CD8+ T cell) immune responses. Preclinical studies in melanoma models have demonstrated its ability to inhibit tumor angiogenesis and reduce tumor volume and metastasis without significant systemic toxicity.
