Overview

KLH-DBI is an experimental therapeutic vaccine designed to induce autoantibodies against acyl-CoA-binding protein (ACBP), also known as diazepam-binding inhibitor (DBI). Extracellular ACBP/DBI acts as an autophagy checkpoint, and its neutralization has been shown to prevent non-alcoholic steatohepatitis (NASH) and suppress NASH-induced hepatocarcinogenesis. The vaccine consists of the DBI protein conjugated to the carrier protein keyhole limpet hemocyanin (KLH) to enhance immunogenicity. In preclinical models, KLH-DBI-induced autoantibodies retard the cell cycle, suppress cell proliferation, and favor ferroptosis in cancer cells, suggesting a potential therapeutic strategy for treating NASH and preventing its progression to hepatocellular carcinoma (HCC).
