KLR-003

KLR-003 is an investigational, oral small molecule inhibitor of Tyrosine Kinase 2 (TYK2) developed by Klear Bio. It is designed for the treatment of immune-mediated inflammatory diseases, with a primary focus on psoriasis. TYK2 is an intracellular signaling kinase that mediates the signaling of pro-inflammatory cytokines, including interleukin-23 (IL-23), interleukin-12 (IL-12), and Type I interferons (IFN). By selectively targeting TYK2, KLR-003 aims to block these pathways while minimizing the inhibition of other Janus kinase (JAK) family members (JAK1, JAK2, and JAK3), potentially offering a more favorable safety profile than non-selective JAK inhibitors. The drug is currently in Phase 1 clinical development, with studies evaluating its safety, pharmacokinetics, and pharmacodynamics in healthy volunteers and patients with psoriasis.
