Overview

KLS-3021 is an investigational oncolytic virus therapy being developed by Kolon Life Science for the treatment of solid tumors. The therapy utilizes a vaccinia virus-based platform engineered to express three genes—PH-20, sPD1-Fc, and IL-12—to enhance its anti-tumor activity. Mechanistically, KLS-3021 combines interleukin-12 (IL-12) stimulation with programmed death-ligand 1 (PDL1) inhibition to promote immunologic cytotoxicity and stimulate cell death in tumor cells. The drug is administered intratumorally and remains in preclinical development as of late 2024[1][2][3].
