Overview

KM-668 is a small molecule inhibitor of Fatty Acid Synthase (FASN) currently under development by Kainos Medicine, primarily for the treatment of Non-alcoholic Steatohepatitis (NASH). FASN is a multi-enzyme protein that catalyzes the final step in the de novo lipogenesis pathway, converting malonyl-CoA into long-chain fatty acids. In NASH, overactive lipogenesis leads to excessive lipid accumulation in the liver, triggering inflammation and fibrosis. By targeting FASN, KM-668 aims to reduce hepatic steatosis and mitigate the progression of liver damage. The drug has demonstrated efficacy in preclinical models of metabolic disease and is being evaluated for its potential to treat other lipid-related disorders such as obesity.
