KM10544 is a novel small molecule inhibitor of interleukin-1 receptor-associated kinase 4 (IRAK4), a serine-threonine protein kinase that serves as a key messenger in Toll-like receptor/interleukin-1 receptor (TLR/IL-1R) signaling pathways. These pathways are implicated in immune responses, inflammation, and various cancers, particularly those with myeloid differentiation primary response 88 (MYD88) mutations. Developed initially by Kainos Medicine, Inc., a South Korea-based clinical-stage biopharmaceutical company, KM10544 has demonstrated preclinical efficacy by suppressing proliferation and inducing apoptosis in cancer cell lines including acute myeloid leukemia, acute lymphoblastic leukemia, diffuse large B-cell lymphoma, and Waldenström's macroglobulinemia, with minimal toxicity to healthy cells.
