Overview

KM10544 is a novel small molecule inhibitor of interleukin-1 receptor-associated kinase 4 (IRAK4), a serine-threonine protein kinase that serves as a key messenger in Toll-like receptor/interleukin-1 receptor (TLR/IL-1R) signaling pathways. These pathways are implicated in immune responses, inflammation, and various cancers, particularly those with myeloid differentiation primary response 88 (MYD88) mutations. Developed initially by Kainos Medicine, Inc., a South Korea-based clinical-stage biopharmaceutical company, KM10544 has demonstrated preclinical efficacy by suppressing proliferation and inducing apoptosis in cancer cell lines including acute myeloid leukemia, acute lymphoblastic leukemia, diffuse large B-cell lymphoma, and Waldenström's macroglobulinemia, with minimal toxicity to healthy cells. Emmaus Life Sciences acquired exclusive rights to its intellectual property in 2021 for oncology applications, particularly resilient lymphomas, and plans in vivo studies for disease selection followed by IND-enabling work, potentially seeking orphan drug designation.[1][2][3][4][5]
