KM114 is a monoclonal antibody targeting CD44, developed as a potential therapy for immune thrombocytopenia (ITP). In murine models of ITP, KM114 successfully ameliorated the condition, demonstrating efficacy at significantly lower dosages compared to intravenous immunoglobulin (IVIg). Its mechanism of action involves the inhibitory IgG receptor FcγRIIB, which is required for its therapeutic function, similar to IVIg. However, unlike IVIg, the Fc portion of the CD44 antibody is not essential for its ameliorative effects. KM114 is being explored as a very-low-dose recombinant therapeutic option for human ITP.
