KM501 is a bispecific antibody drug conjugate (ADC) targeting human epidermal growth factor receptor 2 (HER2), developed by Xuanzhu Biopharmaceutical. The antibody component of KM501 can simultaneously bind two non-overlapping epitopes of HER2—ECD4 (the trastuzumab-targeting domain) and ECD2 (the pertuzumab-targeting domain). This dual binding enables multiple mechanisms, including dual HER2 signal blockade, enhanced antibody-HER2 binding, increased clearance of HER2 from the cell surface, and improved antibody-mediated antitumor activity. The drug exerts its effects through mechanisms such as HER2 antagonism, tubulin inhibition via its cytotoxic payload, induction of antibody-dependent cellular cytotoxicity (ADCC), and T lymphocyte stimulation. It is being investigated for the treatment of solid tumors with HER2 expression or mutation—including breast cancer, colorectal cancer, gastric cancer, ovarian cancer, endometrial carcinoma, gastroesophageal junction adenocarcinoma, non-small cell lung cancer with HER2 mutation—as well as other advanced malignant solid neoplasms. As of early 2024 it is in phase I clinical trials in China[3][5][7][8].
