Overview

KM801 (also known as KY1066) is a fully human monoclonal antibody that targets Dickkopf-1 (DKK1), a secreted protein that functions as a potent antagonist of the canonical Wnt/β-catenin signaling pathway. DKK1 is frequently overexpressed in various malignancies, including multiple myeloma, prostate cancer, and lung cancer, where it contributes to osteolytic bone lesions and promotes an immunosuppressive tumor microenvironment by activating myeloid-derived suppressor cells (MDSCs). By binding with high affinity to DKK1, KM801 prevents its interaction with the LRP5/6 co-receptors, thereby restoring Wnt signaling. This mechanism aims to inhibit tumor growth, reverse DKK1-mediated immune evasion, and promote bone health in patients with bone metastases. KM801 was developed by Kymab (now part of Sanofi) using its proprietary Kymouse platform.
