KM8927 is a genetically engineered humanized monoclonal antibody targeting ganglioside GM2, a glycolipid expressed on the surface of certain cancer cells. It was designed to exert antitumor effects by mediating antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC) against GM2-expressing small-cell lung cancer (SCLC) cells. In preclinical studies, KM8927 demonstrated higher ADCC and CDC activities compared to earlier antibodies and showed antimetastatic activity in mouse models, inhibiting the formation of liver, kidney, and lymph node metastases. The antibody was produced with enhanced effector function, classified as a "Potelligent and Complegent antibody," leveraging Fc engineering to increase immune-mediated tumor cell killing[1][3][5].
