Overview

KM966 is a **mouse–human chimeric monoclonal antibody** targeting the cell-surface tumor antigen **ganglioside GM2**. It mediates both antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC) against cancer cells expressing GM2, which is often found in tumors of neuroectodermal origin, including **small cell lung cancer (SCLC)**, **breast cancer**, and **neuroblastoma**. KM966 induces lysis of cancer cells via both lymphocytes and monocytes, and its activity can be enhanced by combination with cytokines such as IL-2, IL-12, interferon-γ, or macrophage-colony-stimulating factor. Preclinical models demonstrate potent anti-tumor effects, including tumor regression and eradication of GM2-expressing cancer cell lines in vivo. Humanized variants with improved effector functions have since been developed based on KM966[1][3][4][5][7][8][10].
