Overview

KMA.CAR-T is an investigational chimeric antigen receptor T-cell (CAR-T) therapy designed to treat kappa-type multiple myeloma. It specifically targets the Kappa Myeloma Antigen (KMA), a receptor found exclusively on myeloma cells in patients with kappa-type multiple myeloma, and not on normal immune cells, minimizing off-target effects. The therapy involves collecting a patient's own T cells, genetically engineering them to express a CAR that binds to KMA, expanding these modified cells ex vivo, and then infusing them back into the patient to selectively kill malignant plasma cells. Preclinical studies have demonstrated potent anti-myeloma activity and selective cytotoxicity against KMA-expressing cell lines. The drug is being developed through a collaboration between HaemaLogiX and Peter MacCallum Cancer Centre and has entered Phase 1 clinical trials for relapsed/refractory kappa-type multiple myeloma[1][2][3][6][7][8].
