Drug intelligence / Profile preview
KMD-120 (also known as EGFR-GST-ulisertinib) is a novel EGFR-targeted recombinant fusion protein conjugate developed by KMD Bio in collaboration with FusionBioTech. It is designed for the treatment of EGFR-expressing malignancies, including triple-negative breast cancer (TNBC) and glioblastoma. The therapeutic construct consists of an EGFR-targeting affibody fused to glutathione-S-transferase (GST), which non-covalently binds to glutathione (GSH)-conjugated ulixertinib (an ERK1/2 inhibitor). This drug delivery system enhances tumor-specific targeting and stability in circulation. Upon internalization into EGFR-expressing cancer cells, the high intracellular concentration of glutathione triggers the selective release of active ulixertinib, which inhibits the MAPK/ERK pathway, leading to cell cycle arrest and apoptosis.
