KMD111 is a novel HER2-targeted drug delivery platform developed by KMD Bio. It utilizes Protein Corona Shield Nanoparticle (PCSN) / Antibody Conjugated NanoParticles (ACNP) technology, consisting of mesoporous silica nanoparticles (MSNs) loaded with the topoisomerase I inhibitor SN-38 (the active metabolite of irinotecan) as a cytotoxic payload. The nanoparticles are coated with HER2-targeting affibody proteins via a glutathione-sensitive GST-GSH bio-linker. This design provides stability in the bloodstream and enables selective, glutathione-regulated drug release within the tumor microenvironment. KMD111 is being developed for the treatment of HER2-expressing solid tumors, including gastric and breast cancers.
