KMRC011 is a novel, synthetic agonist of toll-like receptor 5 (TLR5) developed primarily as a medical countermeasure for acute radiation syndrome (ARS). It is derived from flagellin and functions by stimulating TLR5, leading to the induction of cytokines such as granulocyte colony-stimulating factor (G-CSF) and interleukin-6 (IL-6), which play key roles in immune regulation and hematopoietic recovery. Preclinical studies have demonstrated that KMRC011 enhances survival rates in lethally irradiated mice by promoting cell proliferation, anti-apoptotic effects, and protection of intestinal mucosa. In clinical trials with healthy volunteers, single intramuscular doses up to 15 μg were tolerated and showed pharmacodynamic activity consistent with TLR5 agonism. The drug has also been investigated for its immunomodulatory effects in models of graft-versus-host disease and ulcerative colitis[1][4][6][7][9].
